One sure way to discern the voice of God is to have a relationsip with him. As a simplistic example - take a friend you'd talk to on the phone everyday; you may not see that often, but because you speak frequently, you know the voice. If that friend calls you and she's laughing, crying, angry, confused etc.... you will know the voice because you know the friend, and you've spoken enough times to know what the friend sounds like.
Same thing with God. Spend enough time with you and you'll know when he's speaking to you, whether it's to convict you for something you've done wrong, to nudge you to act on something, to comfort you in times of sorrow etc....

I see some of the flippant answers that you've gotten. I take this question rather seriously. Paul wrote about the war between the flesh and the Spirit and the World and the Spirit, and the devil and the Spirit. It really is a war. So a warrior in Christ has to deal with unseen things all the time.
But how to distinguish all those and the Spirit of God? Especially when you feel the the answer to your problem is especially crucial. If at all possible, be alone with Him. Just you and Him. You lean on Him for understanding, and it's in His nature to reveal things to His children. The knowledge that He's given you coupled with the conviction of the Spirit, guides you to action. That's the kind of action, that is; the action that's done in trust of Him, that has eternal consequences. Those "Works" are the ones that stand the test of fire at the Judgement.
So that's how I can tell, when the conviction of the Spirit of God brings you to knowledge in the will of God; the God of our understanding.

Simple...take what is being said and compare it to what is said in the bible. If it corresponds to the bible and what God has said in there then most likely it is God, if not then most likely it is the enemy. Even the enemy will use partial truths to keep you from God. God has told us that all things can be tested through the bible.

The true voice is always loving.
If it is not loving it is the ego/Satan.
One must be quite careful here because loving means, loving for all involved. Not just those who agree with you.
This is an easy mistake to make.

Am not so holy that I may hear God speaking openly.
If I hear some warning about something to come then I will attribute that to the prayers my mom prays so God may protect me. And not really that God is speaking to me. who am I so God would speak to me? Im just a poor soul who sins more often than not.
If you hear something out of the blue and you cant tie it to anything like a warning please dont take heed, there are interesting creatures who walk the earth and can whisper all kinds of nonsense into those who wish to hear em and those who are vain enough to think its God speaking.
