What are you planning to go by? Car?
Car rules should be clear and spelled out -- like if someone has a new car and wants to ban eating and drinking, or if a rather nervous driver needs silence during heavy traffic or mountain curves.
If possible, try for a little extra room in the cars, rather than packing them to their limits. If a car breaks down, then the others can pack into the other cars.
Plan for regular breaks during the trip -- to stretch, bathroom break, and just relax.
Make sure the same people go back to the same car -- or organize a way to have a real head count before you take off from each break spot.
Set limits on the luggage each person can bring. And the stuff each person can bring back!
It might be best to rent a bus and a driver for that many people, though. It'd probably be cheaper gas-wise, and people can have fun instead of worrying about driving. Depends on how wild of an area you want to drive to.

Among other things (I'm sure you'll get lots of different answers!), you need to transport all their gear too... Don't forget the can-opener and the matches.
