I don't know about getting on the NY times best seller list but I do know that if you are not a published author many publishing houses will not even look at your manuscript. Agents today want to represent only published authors. So the only way to get published is to do it yourself. There are many self publishing web sites that will help you do this and get your book into the stores. If your book sells out then you reorder more. Who knows maybe you will be the next Robert Ludlum.

Get an agent to market your book. You'll have to start with the 'Great Query' letter to get their attention, have a well written grabber, know your target market and your competition, do your homework on the genre you are working in, and be able to sell why your book is different, interesting and worth the purchase price.
