There is this minister would in marrying this couple the groom is the minister's best friend. The groom is cheating on the bride to be. What should the minster do?

The minister should talk to the groom, and find out what is going on, and remind him of the covenant of marriage.
If I were the minister, I would not marry them until this problem is solved. In my Church, before the pastor marries a couple, they must go through marriage counseling.
It is not the pastor's position to tell the bride.

If the minister knows of the cheating, he should refuse to participate in the wedding. He should also inform the bride. He has a responsibility to protect his flock.

Assuming the minister knows of the cheating, he should perhaps recuse himself from the ceremony and ask that the couple find another minister.
