Your question was "only breed of dog to be developed in the US." And yes there are quite a few. Off the top of my head the "American Cocker Spaniel" is one. There is also the American Water Spaniel, American Staffordshire Terrier, and the American Eskimo Dog. These breeds have "American" in thier breed description to deferentiate from the "English" variety.

Someone mentioned the coonhound and yes, the Black and Tan Coonhound was also developed here in the US. My information comes from "The Complete Dog Book". They say "The Black and Tan Coonhound is one of the few truly American breeds. Developed in the Deep South by hunters who blended the unique traits of several early hound breeds." This suggests that they did not originate here, but were bred here to create this unique breed. Still it's origins lie somewhere else.

There are many examples of breeds developed and refined in the US. The American Eskimo dog is a Nordic breed and descended frm the European spitzes. The spitzes originated in Europe, Germany and Italy. They were brought here, where they were bred and thier standard refined. But they did not originate here. This is the case with most of the breeds.

Without sitting down to read the entire AKC book, which is quite large, I couldn't tell you if there was one breed that was found only in the US and bred to a standard here, then accepted into the American Kennel Club. But many, many were bred and/or developed as you say, in the US.

There are actually several breeds of dogs that were developed in the USA; many states actually have their own "Official State Dog", of a breed native to that state. Just in AKC-recognized breeds alone(and you can go Googling for each breed I name here, to find out more), there's the Boston Terrier(Mass.), the Chesapeake Bay Retriever(VA and MD), the Alaskan Malamute(AK), the American Water Spaniel(Midwestern US), the American Foxhound(VA), the American Cocker Spaniel(US in general), the Australian Shepherd(yes, it was developed in the US by Basque immigrants), the American Staffordshire/Pit Bull Terrier(US in general), American Bulldog(Southern US), Redbone Coonhound(Southern US), Bluetick Coonhound(Southern US), Black-and-Tan Coonhound(Southern US), Walker Coonhound(sometimes synonymous with American Foxhound-Southern US), English Coonhound(yes, another American creation-no raccoons in England-Southern US), Plott Hound(NC), and Toy Fox Terrier(US in general). There are also several breeds, primarily bred as working/hunting dogs, that were developed in the US, which are not recognized by the AKC, and these include the Louisiana Catahoula Leopard Dog(State Dog of Louisiana), Southern Blackmouth Cur(TX), Blue Lacy(TX), Florida Cur(FL), Leopard Cur(Southeast), Mountain Cur(Southern Appalachia), Treeing Feist(TN), Tennessee Treeing Brindle Cur(TN), Boykin Spaniel(State Dog of SC), and the oldest and most natural of the native US breeds, the Carolina Dog or North American Dingo(Southeast). Note that word "Cur" here does NOT mean a mixed-breed dog, but a working breed that typically developed from the Spanish Conquistador's Mastiff-type war dogs breeding with the Native American's dogs of the region, then later with an infusion of European hunting/guarding breeds. Information about the "Cur" breeds can be found below, as well as information on the Carolina Dog. Other breeds can be researched via the AKC or UKC websites.
