There's couple of ways to say each one, depending on where you are and which level of education you're talking about.
The general form is: Cual es tu clase preferida?
For "clase" you can also use "curso," "estudio," or "materia."
For "preferida" you can also say "favorita" or "predilecta."
The response would be:
Mi (clase, estudio, materia) (favorita, preferida, predilecta) es.."
