There is one way if you can handle the kitten without offending the mother cat. It is not fool proof but it works about 50% of the time. Usually where the sexual organs will be (under the anus) if the shape is circular it is a male and more a triangular shape it's a female. My Grandmother use to do it an she was 100% accurate but she had done it for a long time.
Best of luck but really you will know as the sexual organs start maturing in 6 to 8 weeks will be pretty much determined.

It is difficult without comparing a male and a female. Just stop by a vet and they will tell you in a second for free. My kitten is now 4 months old and I can finally tell absolutely.

By this age you can tell. You can tell from the minute they're born if you have a boy or girl. I have 7 week old kittens, and I could tell from the minute they were born that they were all boys. If the momma cat will tolerate it pick them up briefly and check for a set of nuts. My momma cat trusted me to pick her babies up right away. We tried not to pick them up much before their eyes opened so they wouldn't get scared.

It is easy to differentiate the sexes once they are born
The male pouch is a bit bigger and looks triangular
compared to the tiny almost hidden of females
I always announce the numbers of males and females to my siblings each and every time a litter is born

At three weeks old you should be able to tell. If she doesn`t have those two little buldges she is a female and she will stay that way.
