I'd start in Times Square to get a feel for the place. I'd then hop on a boat and circle the city(see link below). You can see a lot on a two hour walk through Little Italy and Chinatown. Have a great dinner and then drinks at the Oak Bar in the Plaza Hotel.
Feel free to substitute a trip to a great museum like the Metropolitan Museum of Art or the Museum of Natural History. Broadway shows are great but expensive. If your a shopper hit Fifth Avenue between 50th and 57th
Count on spending a minimum of $150(not including hotel or shopping) to do it right.

Check out the Metropolitan Museum of Art on 5th Av, take a cab to the Empire State building, and go to the observation platform, then head west and a little north to check out TImes Square. From there take the B/D, or the A/C line subway to the Village (West 4th St stop), and walk around. Hang out in Washington Square Park. Explore a little. In the daytime. U like pizza? The BEST is on Spring Street in SoHo, and its called LOmbardi's. If you want to go to where the WTC once stood, from Times Square, take the C train to Chambers St, and say a prayer. Seriously, there are SO MANY things to do there, its hard to pick even just a few. Most important is to walk around when you can, just to get the feel of the City. Dont forget to have a Sabretts hot dog from one of the vendors....

start from MET. I heard they just acquired the famous "snake charmer" by Jean Leon Gerome. Here is the link.

Then head to the natural museum at 82st and central park west.
Then go downtown to the Metropolitan museum and see the Van Gogh's "starry night" and "women picking olives" (among others) and many other famous pieces by picasso. Student get in free.
After that, go see "Marie Antoinette".
