It is a command given by G-d to Abraham and all of his male descendants throughout all of their generations. (Gen 17) The significance is that it is an act of faith an obedience on the part of the parents (at 8 days old, the boy doesn't have much choice in the matter).

It also is an outward sign of the circumcision of the heart (Deuteronomy 10:16 & 30:6). That is to say, a heart that is open to G-d. Adam was created in a state where his heart was completely open, but it wasn't a "circumcised heart" since that would imply that the heart was previously uncircumcised. When Adam sinned, G-d gave him a coat of skin (Gen 3:21) meaning he became uncircumcised both in the heart and in the flesh.

It is a symbol that he is part of the chosen people of Israel. It is an indication of being part of the covenant that God made with God's people.
