I remember visiting a Sinagogue in the past that a man was calling to say an special prayer. I heard that was because he was a cohen. But I never knew what make him a cohen. I would like to know who is considered a Cohen in our days?

a "cohen" is the preistly class from biblical times. they have a tradition from father to son as to their tribal status. one cannot become a cohen; either one is born a cohen or one isn't. in many synagouges, there are no cohan's, and therefore certain prayers -- reserved for a cohan -- are not said!
