In this verse Jesus is talking about pride. Someone who is rich in spirit is rich in pride. Someone who is poor in spirit is humble and recognises their own sinfulness, and by extension their need for salvation.

To be poor in spirit means to realize that you are not "all that and a bag of chips" so to speak, but that we all have sinned and fall short of the glory of God. When you humbly acknowledge that and accept the gift of salvation Christ paid for you, you become a citizen of the kingdom of heaven, your eternal home when this life ends.

To be poor in spirit means to not know where to turn, but to know that man does not have the answers. Please use the following link to read a great story that illustrates this point. I'm trying to only use one paragraph, or I would quote it here. It starts in Alma 31, but 32 has the significant part. Poor in heart = poor in spirit.

I was driving to work this morning and saw a homeless man. I gave him a few dollars and asked if he knew Jesus. He said yes, that Jesus was His savior and he wouldn't even be alive without Him. As I drove to work, I thought, there are people sitting in million dollar mansions who don't know what that man knows. As a matter of fact, every single person I've run into on the street is acquainted with Jesus Christ. And I don't think they are just saying it, either. That is what poor in spirit is....when you have nothing else, and you know how much you need Him.

Poor in spirit means being humble before God and mankind as well. Living life in recognition that we are called to be a servant to all... and a ruler over none. Poor in spirit denys exaltation of self and gives all of ther glory for anything that is accomplished through him or her to God.

Pride was the reason Adam & Eve lost heaven.

Humility, or being poor in spirit, and trusting only in God for sustenance, gains heaven.

Those with the greater spiritual need are more likely to perceive their need and depend on God alone and not their own goodness. Paul notes the same prinicple in Romans 9:30-31.
