I am saying this informally, so I do know that I use "tu." I am just confused on which verb to conjugate - "ser" or "estar" .... ?

¿Qué está en tu casa? would be a direct translation, although I don't know that it would come off very natural.

You didn't elaborate much on your question. The context might change the question a lot.

¿Qué tienes en tu casa? ¿Qué tienes en la casa? ¿Qué hay en tu casa? are all subtly different ways of asking.

As far as ser vs. estar.... Estar indicates physical location (i.e. "what objects are phyiscally located within your house?"), so you should definitely use it over "ser" in this situation.

¿Qué hay en tu/su casa?
or
¿Qué es lo que hay en tu/su casa?

use 'tu' for a colloquial conversation
use 'su' for a more respectful tone.

I think you'd use estar (if I'm not mistaken, it denotes location).

never say: What is your house?
It's where is you house'?
¿Donde esta tu casa??
