Depends on the choir. Usually, you want to approach the choir director, or whoever's in charge--I usually do it at the end of the service. Then, they'll either schedule an audition (if they're snobby), or just tell you when and where practice is.
If you're really good you may get drafted by existing choir members.

Ask if there are auditions. At my church, there aren't any auditions. Anyone can join and sing.
