Not longterm...but focussing may be diffcult if you have long or short sighted vision. Your eyesight is pre-determined but can be corrected via laser.

If you make a habit of it you are overworking your eyes, which may cost you in eyesight later in life, as your eyes naturally become weaker. Why/ Because of the motion of the vehicle causes you to constantly refocus, and that does wear and tear on the tiny little muscles the work lens to maintain focus. B.

No, but you may become motion sick because of being in a moving vehicle.

The moving vehicle is moving making you or the book move so it makes you look from the corner or whereever.
