It is a Biblical expression (2 Corinthians 11:19). In this verse it is used in the positive sense, as the Corinthian Church is prepared to suffer fools gladly. To understand its meaning, one has to be aware of the background. Paul is using irony. The Corinthian Church has turned away from his authoritative teaching and is listening to all kinds of half-baked ideas from ill-informed people. They are, in other words, all too happy to put up with stupid people, while rejecting him.

Nowadays it tends to be used in the negative sense. "I don't suffer fools gladly" means that I am intolerant of people who are stupid, I am irritated by foolishness.

Personally, I take it to mean that people who try to tell me how it is without sufficient back up are to be ignored. That's quite simply ignored, it isn't necessary to point out that they don't know what they're talking about.

Sort of like, don't be around dull annoying people and take it with a smile. Turn around and leave, don't hang around fools.

You are not happy to have to deal with stupid people.

Usually it's "i don't suffer fools gladly". Go and look up the definition of each word. Then piece it together.

"For ye suffer fools gladly, seeing ye yourselves are wise." II Corinthians 11:19.
I take this to mean that we should not be smug thinking we know it all.
