I think just "happy anniversary" would be fine. Anything more than that would clutter up the cake. Write your sentiments in a card, or better yet, tell them how you feel in person. It will mean much more than seeing it on a cake that will be cut up and eaten.

cake should have minimal writing on it, as then the guests especially the children start hankering for which alphabet they want etc. so keep it simple like : 25th or happy 25th or just happy anniversary. u could try writing it out in silver though.
