the question is a little open ended, so here some answers to fill in the gaps:
1 in boxing, there are several places considered key and can result in a K.O. the jaw, the temple, square on the chin or even a straight cross smashing on the nose and right between the eyes. Down below, an uppercut to the solar plexus, or a hard hook to the liver will drop even the most seasoned professionals (de la hoya, vargus, etc..)
2.In no holds barred, a good punch to the dong will drop em quick

If you have a good hook, go for the ribs so he can open his arms to allow hits on his face. Its a strategy i use in boxing.

Or if you can keep up giving your opponent frequent hits without stop, hit his face, although he will likely be blocking, the frequent hits will end up making his arms go down. then go for the nose, make his eyes tear up.

Oh yeah, if he has a soft stomach, hit his stomach. Most boxers dont, so watch out.

if u want to do permanent damage, then the nose is best, you should use an open palm to get best surface area contact, be sure you use a motion that comes from under to pushing upward. If you just want to get a KO then point of the jaw or liver. If no holds bard, the jewels is good to stop any guy.

it all depends. if theyre a trained fighter, hits to the face, nose, jaw, ribs, whatever, wont do much damage at first. their endurance is so built up, i mean come on, they do this for a living. but under any other curcumstances i would suggest a nice, clean, hard shot to the chest. itll send them back, knock the wind out of their lungs, and have them at your mercy for at least another 10-15 seconds.

Square on the nose. The eyes well up with tears and they can't see to defend from the next slew of punches you throw at them. If you're lucky, their eyes will swell shut a little as well.

if your talking about ending it quickly hit them as hard as you can on the bridge of the nose or a quick shot right on the temple.....lights out
but remember this should be done by a trained professional not done in the privacy of your own home...
