What is that song about? I understand the words, but can't figure out what it is about. Does it have to do with religion?

It is indeed very hard to "Get" ,unless you are from Colombia, i guess. I'm a native speaker and i'm still not sure.
Oh well, what i understand is that Carlos Vives and a few other guys were going to sing on Saturday for the Virgin. One of the guys (Moralito) thought that he could sing/play (some musical instrument) better than Carlos V. Once Moralito hears Carlos playing (le callo la gota fria) literally meaning that a cold drop fell on top of him , But what i think he relly means is that he was speechless, amazed , that it dawned on him how much better Carlos could play.

The Killers sang some Spanish in "The Ballad Of Michael Valentine" and "Happy Birthday Guadalupe" Yeah, it's good

Wow! That's a weird song. You need to get a real Colombian or Venezuelan to answer that. I'm from Spain and that song to me is in Chinese for all I know.

One thing I CAN tell you it's got NOTHING to do with religion!!!

The reference to Mary is that traditionally Saturday is dedicated to Mary, that's all, but it's just accidental to the whole song.

I think its just a folk thing. BTW, "vaina" in local idiom means "thing" ! ☺

It makes reference to the virgin mary in a single line, but I don't find any more religious references in the song.
