There are two routes to go to Velankannai from Chennai. You can take East Coast Road, which goes via Mahabalipuram, Pondicherry, Cuddalore, Sirkazi and to Velankannni.
Another route is take National Highway 45 till Tindivanam. After Tindivanam, a state route takes a turn towards, Neyveli, Kumbakonam and to Velankanni.
Both routes are good. It depends on the location where you reside at Chennai. If you are residing at South Chennai, ECR (ease coast road) is the best. If you located elsewhere in Chennai, then take NH. Hope this helps you.

You have to take the southeastern road out of Bangalore toward Hosur. Then you continue through Sulagiri, Krishnagiri, Uttangarai, Chengam, Pachchal, Tiruvannamalai, Tirukkoyilur, Panruti and Nellikuppam. This will get you to Cuddalore in the north of Valankanni.

Bangalore-Hosur-Palaccode- Dharmapuri- Salem-Namakkal- Tiruchirapalli- Thanjavur- Tiruvarur-Nagapattinam- Velankanni. About 470 Kms.
From Hosur to Dharhapuri you can also go via Krishnagiri
