If you are convinced it is a serious threat, then call right away. If they, by any chance do not take YOU seriously, then exaggerate your fears a bit.
Whatever you do, DO NOT call the police if you think the threat is not intended to be serious. You can get in trouble for filing a false police report. I'm NOT trying to talk you out of it. Just be sure, so that when the police respond, you can still be sure, and communicate how serious you take the threat.
Think of this. Many robbers threaten people with a toy gun. There is absolutely no danger from the toy gun, but if the victim BELIEVES the gun is real, the police will treat the crime as a genuine armed robbery.
Be very sure, and be very convincing.

Much depends on how you assess the threat. If you think it's credible and the other person is capable of carrying it out, and it wasn't just careless talking in the heat of agitation, then it may make sense to call and make a report.
There are two reasons for making a report of this type. The most common is to document the fact that someone threatened you - to put it on record in the event something more happens, an attempt is made, or the anger escalates. It's a benefit to you to establish a record of happenings. Another reason is if the threat crosses the threshold of just words, and gets into an actual violation of statute for your state. The seriousness of the report and the evidence you may or may not have to corroborate it, may make a difference on how much the police will be able to assist you.

Depends on how serious the threat is, how frightened the threatened is, what proof of the threat or witnesses are available, etc. etc.
If it's a serious threat by someone who seems to have the ability and intention to follow through, I'd say the sooner the better.
