Some of the answer depends on your age and what sports you are attempting to condition yourself for, but the short answer is run -- if you have ever played a coached sport then you know that conditioning comes from increasing your aerobic ability. Treadmills, steppers, etc. will help somewhat but nothing beats getting out there and running.

The type of running you do can also aid in the sport that you are playing as well. There is a reason that basketball coaches run sprints and stairs because you need quickness and endurance. Football players run in their equipment to get used to carrying the extra load.

So buy yourself a good pair of shoes and get out there and run...

Endurance has to do with the heart and the lungs, is it your breathing? If you get winded you'll need to brush up on your cardio work. Start run/walking, riding stationary bike, do you belong to a gym?
Do your muscles get too tired? Strength training will help this area. Knowing the sport would help to know what to advise you with.
