macbeth's wife was haunted 4 the murders she was responsible. she falls ill & starts hallucinating abt her hands being covered with blood..so she keeps rubbing her hands & washing it.in context to the situation she says the above sentence.the "damn spot" refers to the stain of blood she assumes to be on her hand.

if i am right, this is the part where the crime has already taken place and Macbeth's wife knows about it and her conscience will not leave her alone, therefore she is paranoid, going onto crazy and in her mind she can see the blood-which means that she was implied-which plays a big role, therefore her consience is haunting her for the deed she did.
