In the first paragraph you should introduce yourself and explain how you learned of the position. Your next paragraph should highlight some of your past experience as it relates to the position. Your third paragraph should relate to the future at the position and set up for a follow up - where you can be reached, etc. The cover letter should not rehash what is on your resume - perhaps you could use it to talk about a success you had that isn't on your resume that relates to the position you would like. I assume you mean "cover letter" that you put with a resume? Typically the best things to mention in a cover letter are a little about yourself, your interests, why you're seeking that particular job, why you would be good for the position, the skills you possess that qualify you for the position, your career goals, your experience that has brought you to the point of being the right person for the particular position to which you are applying, etc.
Employers like to know why you want that particular job in relation to your career goals. They also want to hear why you think you qualify before they start the grueling task of checking your work experience and references.

if you mean for a job application, i usually just write,
dear sir/madam, i am writing in response to your advert for the position of ....... ?advertised in .......?. please find enclosed a copy of my cv and details of references. thank you . yours faithfully........
