Its a phrase that comes from online gaming. It is a reference to newbie which means you're new to the situation (unlike those pro 15 year old hax0rz) Newbie became n00bie which is now just noob.

If you call ppl noobs then they won't think you're a noob so use it freely.

A really lazy guy, trying to be "cool," thought having to type 4 different letters in an already shortened word was just too much work. Thus, the word noob was born.

n00b is a corruption of the word newb or newbie.

A Newbie would be a new user, a neophyte. A n00b is someone on the internet who is pretty much childish, irritating, stupid, offensive, and a troublemaker.

People assume they are synonymous, and they are NOT. newb(ie) has no negative connotation whatsoever, it's not an offensive title, n00b however, is negative and is meant to be demeaning.
