If subway is the fast way, please give me specific instructions. But, to do the trip by street level is preferred. Don't like to waste time. Without worry about cost.

Take the C train uptown From Chambers Street (at
Church Street) or from Broadway / Nassau Street to 81 Street.

Without worrying about cost?
Cab. Thats easy.

There are three lights on the top of a cab, look for one where only the center is lit, stick out your arm and he'll stop for you.
If no lights are lit on the roof, he has a fare.
If the two outside lights are lit, hes off duty.
If all three are lit, hes about to end his shift and may or may stop for you.

The fare should be about 12-15$ probably a bit less.

Nobody will be able to give you exact subway instructions without knowing where you're starting from.

There are express busses, but they will take much longer than the subway. I believe it is the C train that has a stop right under the museum. Check out New York City Subway maps.
