It is just a stronger statement than No Smoking. It would be like putting an exclamation mark after it. Perhaps there is a danger (like the presence of oxygen, gasoline or something like that) that makes it not only unacceptable but down right dangerous to smoke in that area.

Positively is an adverb it emphasises that there is no smoking.
They can use force to stop you from smocking , physically throw you out, prosecute you have you thrown in jail, law permitting(in an air plain).

You cannot smoke at all, even in the wash rooms, cos there are smoke detactors.

they do 'negative no smoking' twice (i.e they smoke twice). two negatives make a positive so it becomes 'positively no smoking'
