Charlotte is a straight shot right down I-85 from Burlington. In a car at normal speeds, it's about a 2 1/2 hour trip. Man, I hope you aren't planning on doing that every day. Traffic is crazy, and it seems like the construction crews are always messing something up. If things are moving along too well, somebody has to wreck their car or truck.

Straight shot down I-85 - you could take NC 49 as a scenic route to avoid I-85 when it is backed up.
