A runway names "27-09" can be approached for landing by heading 270 or 090 on the compass, or, in layman's language, the runway can be used by lining up with the runway at an appropriate descent and then heading 270 or heading 090 as it runs exactly east-and-west.

Airport runways are designated by a two digit number equal to their magnetic heading in degrees divided by ten and rounded. A runway with a double designation can be approached in either direction. Thus a runway designated 09/27 runs east and west relative to magnetic north.

The numbers represent the compass headings, one for each approach direction, so the numbers are always 180 degrees apart.

For simplicity, the last zero is dropped, so your example is for a runway running exactly east-west at 270 degrees or 090 degrees, with 000 degrees being due north.
