For a tourist, First piece of advice, DO NOT RENT A CAR !
Get a Cab or use public transportation.

20 Min off peek in a cab (at 3am with no traffic and a good driver) 90 in rush hour on a cab. 45-60 min on a city bus.
With a lot of luggage, or kids or both, get a cab.
Ask your hotel, they may have a deal with a car service, or other hints. Don't come into NYC without reservations at a hotel.

From personal experience 20 mins. 45mins the most with traffic. I would rather stay at a hotel in Manhattan than in Queens. Good Choice!!!!

On a good day (little traffic), it could take about 20 minutes to get to The Plaza on 59th ST and 5th Ave. If there is a lot of traffic it could take an hour (or longer).
