Log in as Administrator

Start>Control Panel>Folder Options

Select the View tab and uncheck the 'Use Simple File Sharing' checkbox, it's the last one in the list.

Set up a new user with a password

Find the folder that you want to protect, right click on it and select 'Properties'

You will now have an extra tab called 'Security'

In the top box ensure that only your new user and possibly Administrator are present

Highlight your new user in this part of the screen then in the lower half tick the box that says 'Full Control'

Do the same for your Administrator account if you want them to have access also.

Click OK and now only the users that you have designated will have access to that folder.

There is a free program available to do this. It is available through www.download.com and is called Folder Access. you set a password to access the program, and the program must be accessed to lock and unlock the selected folders.

With a program, like this one:

http://www.password-protect-software.com/

You can't do it without some type of program (as in, there's nothing built-in to Windows to accomplish such a feat). And I can't seem to find any FREE programs that do this either. They all want money.
