Generally speaking, when you kill someone intentionally, you have committed murder. When you accidentally or negligently cause the death of someone, you have committed manslaughter. They are different crimes, as a function of intent, and murder usually carries a more harsh punishment.

All the above will vary by state. A lawyer will explain this better, but I'll try lol. Murder is more of a hands on I guess, where as manslaughter is more of a negligent-ie: dui or driving at an excessive speed causing an accident kills another driver=manslaughter. That's pretty much the jest of it, like I said a lawyer or such can explain it much more detailed.
Punishment will depend on a lot of factors, including the state, previous criminal history, circumstances involved in the case, etc.
Murder is worse. Most states also have different degrees of both murder1,2,3 manslaughter 1,2 involuntary manslaughter.

The difference between the two crimes lies in the INTENT of the criminal.
Punishement definitely differs, but murder does not necessarily get harsher punishments or otherwise. (Other factors also play a role is meting out punishment).
As a rule, I would not say one is worse than other. It all depends on the facts ove every individual case.

The difference is in intent. Murder is when you intend to kill or at least cause major injury. Manslaughter means that the death was an unintentional consequence of your actions.
Manslaughter has lighter punishments, less time in prison and at lower security prisons.

I believe the difference between the two is manslaughter is when you make the person suffer(beat and abuse) before you kill them and murder you kill them instantly. Which is worse?! Well, I would rather die instantly than go through the abuse. I don't know why or understand why there are different in punishments.
