They're sacred to different religions because they mean something to the religion.
The Holy Land is where Jesus was born and lived, making it sacred to Christians and it is sacred to Jews because it is the promised land to them.
Temple Mount/Dome of the Rock is sacred to Jews because it was the site of the first and second Jewish temples and it is also significant to Muslims because it is the site of two major Islamic shrines there.
Mecca is the holiest place in the world in Islam. The well of Zamzam is located there, and water from this well is believed to hold health benefits and special properties.
Lhasa, Tibet, is the capital of Tibet and where the Dalai Lama holds his seat.
The Virgin Mary was sighted at Lourdes, making it a sacred sight for Christians. People who visit Lourdes are said to receive miracle cures if they are ill.

A place can't just be designated as a holy site for a religion. There is usually a pretty significant reason as to why it's so important to a religion.

These places hold a spiritual meaning to people of that particular religion to which it is special. Like Christians go to Lourdes if they are ill, it is because they have faith in the Virgin Mary.
Everybody has their own beliefs and everyone has a place they hold specially in their heart, regardless of whether they are religious or not.

I hope that helps.

There are real forces at work in Creation. The #12 symbolizes
12 vortexes at work on our world. One is said to be in the
Bermuda Triangle, where Atlantis placed a giant crystal that
amplified the power. Ezekiel 1:22 KJV.
The only thing about me, is I don't approve of those who talk
about sacred this, and sacred that, and then hate on a man.
What is more sacred, a material place, made of spirit?
Or a human being with the spirit of God in it.
The Book says if one says, "He loves God, and hates a man,
then he is a liar!"

There is said to be an Native American Burial ground just down the street from me. They built a building on it to sell furniture. I went in once, long before I knew it was a burial ground, and got a very uneasy feeling. So left. In the last 20 years every business that has tried in that spot, on a very busy corner, has failed.
