Grammatically, "How may I help you?" is correct, but I think either one sounds equally polite.

The very first thing out of your mouth should be the name of the company, your name then - how may I help you.
ABC Company, this is Angelfood speaking, how may I help you today...less formal - start it out with a Good morning, afternoon, evening etc. Don't talk too fast, don't eat or chew gum, don't sound like a recording and BE NICE!

I think can over may. Can sounds more positive and affirmative. As in I can and will help you. May sounds like, Well I might be able to help you, let's see.

how may I be of assistance?
may over can .... because 'how can I help you" means that you have the ability and resources to help ... and sometimes you may not be able to resolve every problem brought to you. Some of them will be out of your control.
