It has more to do with sedentary lifestyle. Try to put 15-30mins of a good workout in the morning or early evening into your routine. Focus on some good glute excercises. There are also some websites that give "office chair
excercises".

Some people often talk about tightning and releasing the glutes in reps of 20 every two hours while seated. I prefer to save the workout for when I'm at home, and I try to do little things that add to my activity, like...save the last 10 mins of lunch for a quick walk - park further from the office adds walking too, and I try to do as much running around the office as I can throughout the day.

I highly doubt that can cause your butt to flatten? Because I sit on mine all day at work as well and i'm doing fine. If you feel yrs is getting flat or saggy or whatever the case may be, you should try lunges at home in your spare time. It's a great workout to tighten your buns!

take breaks every half hour, and stretch. you can do it in the bathroom if you don't want people to see you doing a quad stretch at work, lol. But get moving a little bit, and exercise. Eat healthy food, and all that usual jazz you probably hear. Just avoid sitting for all 8 hours straight.

yes, because you aren't using the muscles enough. Flex them while you sit there, get up and walk around at least once an hour, walk on your lunch break and your other breaks. Climb the stairs whenever possible, and (at home) lay on tummy and raise legs up 10 to 50 times each (progressively) every night to build the hamstrings and gluts.
