this is hypothesis on lab assignment that I am having a hard time with.

We are looking at an enzyme substrate reaction. You have a set number of organisms with the appropriate amount of enzymes. The only things that will increase the rate is change is organism number and increase in temperature. Supposing the control and experimental organisms are equal in number increasing the amount of sugar and pyruvate will do nothing except to prolong the the time to fully breakdown these two materials. It has no effect on the rate.

Assuming that they are the substrates for the cells being grown, then they will increase the rate of fermentation until another limiting factor comes into play such as nutrients.
