Ask them about their health policy, ie when parents are expected to keep children home and when children can return after an illness.
What their discipline policy is or if they even discipline at all, some don't.
What they do if there is a child in the room who is bullying or making life tough for other children.
Do they keep a folder on your child and her development and let you know what she is doing.
Who they release your children to if you can't make it, do they use a code word, a special tag or card that the person presents, verbal or written permission for a certain individual to pick up your child, etc.
What do they do in case of an emergency where the center has to suddenly close down or evacuated?
What do they do if you are late to pick up your child due to bad weather or traffic?
What types of meals and snacks do they serve, are they healthy?
Are they able to work around any health problems such as food or other allergies, asthma, etc.
And of course make sure they are licensed, get references, and make sure they have enough staff to take care of the kids, some places don't hire enough people. This happened at my daughter's day care and she ended up getting outside and nobody knew it for several minutes.

I would make sure there is some kind of security system to get into the building. You don't want just anybody to be able to walk in off the street. Also I would check to see if they have a fire alarm system that is monitored by an alarm company or do they have to take the time to dial 911, you don't what them taking the time to call 911 before they your child out. They should leave the calling to the alarm company so you can put your attention to getting the children out of the building. The best thing for me when my daughter went to daycare was the ones that provided food for them, that was one last thing I had to do in the morning.

Make sure you ask the teachers and aides themselves what kind of education and experience they have don't take the supervisors word on it. Also ask about the discipline policy, the number of children enrolled in the program as a whole not just the number in attendance that day. Some other questions are: What is the different age groups of the children? How do the different age groups interact ? What I mean is if the program is say ages 2-5 then do the 2 year olds play with the 5 year old , in the same area as the 5 year olds or in another space altogether. Are the teachers assigned to one group of kids or do the teachers move among the different groups? What is the schedule and how strict is it? Also how much time is spent outside and how are the different ages split up outside or are they even split up ? And what is there for the children to play with outside and do you think that it is age appropriate?

Ask what there curriculum is because the earlier they start to learn, the better. Ask about the teacher-child ratio. the lower the better. Ask how much TV time there is a day. You don't want your little girl sitting around like a zombie all day in front of a TV. See if meals are included in the tuition. Ask if they administer medication because some places don't and you will have to go there to give your daughter meds. Good luck:)

You must make sure that the daycare setting is registered and that the staff caring for your child are fully qualified. Ask for a tour beforehand to make sure you and your daughter are happy with the setting. Your gut feeling as a mother won't let you down.
