Hi there
Usually the culture and sensitivity testings are done together !

Culture is done to know whether a particular organism, bacterial or fungal, is present in the specimen like pus, blood, fluids etc.
Specimen is plated into favourable environments like agar and tested for the presence of the organism after
24 hrs or 5 days, depending upon the technique used.

If there is presence of the particular organism, they are tested against different antibiotics or medicines ..to know, which antibiotic or medicine will act the best upon which organism...! Based upon the result of the sensitivity test, the doctors prescribe the medicine.
example : if an organism S.Typhi, which is causing typhoid is resistant against chloramphenicol but sensitive against drug ciprofloxacin , it means that the drug chloramphenicol will NOT act on the organism , but ciprofloxacin will ...

best wishes..

In microbial sensitivity testing, the offending organisms that have been isolated by culturing are tested for their sensitivty to a range of antibiotics. When plated on nutrient agar plates that are dotted with antibiotic disks, a zone of inhibition of bacterial growth will show how sensitive the particular bacterium is to a given antibiotic. We do this because we need to know which of many antibiotics is the best choice to treat the infection. There would be little usefulness in treating a bacterial infection with an antibiotic to which the bacterium is resistant.

In addition to determining which antibiotic is best to treat the bacterial infection of the patient the pattern of sensitivity and resistance is also used to monitor the "signature" of particular organisms and to identify newly resistant strains. MRSA (Methicillin Resistant Staph Aureus) is a relatively new problem in hospitals across the US. This particular organism is very difficult to treat.
