The penicillin will still work but the great concern will be the irritating effect on your stomach. Penicillin causes gastric upset so if you add alcohol it will make gastric irritation worse, so use it with moderation if unable to avoid drinking while on the penicillin course.
The main concern on drinking acohol is when you get drunk and the tendency is you will forget to take the medication or miss the dose. If you get drunk, you are more susceptible to certain infections and your immune system might not fight your current infection as strongly as normal. So what is the point of taking penicillin. This won't give benefit and on the contrary it will aggravate your condition which will prompt you to repeat the course and if penicillin resistance develops, a higher generation of antibiotics will be required.

Alcohol will make the penicillin ineffective.
The whole goal of antibiotic therapy is to keep a certain level of antibiotic in your blood stream so that it can inhibit the growth of the microorganisms. This is why doctors always say you have to "take the whole course."
If you drink alcohol, you make the antibiotic ineffective, so you don't have this consistently high dose. You end up with a varying concentrations at varying times. This cuts the microorganisms a break and can allow some to develop a tolerance to the antibiotic.
There have been some strains of TB (tuberculosis) that have become drug resistant because homeless alcoholics have continued to drink while taking antibiotics, allowing the TB to develop tolerance to the drug.

It's actually not THAT okay to drink alcohol whilst taking penicillin. Alcohol reduces the strength of the penicillin but not enough to completely void it out of your system. it'll just take longer to get better, depending on what you're suffering from but most often it's just a time aspect.

Why didn't anyone check out the site posted by the first person. No the alcohol has no effect on the penicillin if you are taking certain kinds of the drug it can cause facial flushing.

Most of you are jumping to conclusions which are ill informed. Alcohol will NOT affect penicillin effecitveness.
A few antibiotics should not be taken with alcohol because they interact. Just follow the link to www.gpnotebook.co.uk which leads to this statement;
"There is a generally held belief that alcohol should not be consumed while on a course of oral antibiotics. This advice is correct if a patient is taking metronidazole, cephamandole (may cause a disulfiram-like reaction), and possibly oral ketoconazole (may cause facial flushing). However there is no contraindication to the consumption of alcohol with other antibiotics."
Please get your facts right before giving other people advice!

penicillin is FINE with MODERATE alcohol. Other antibiotics are absolutely not (eg metronidazole).
Penicillins all work fine with alcohol.
Your Dr will say no as its best not to mix EtOH with ANY drug unless you have some medical training.

When you take ANY antibiotics you should never take alcohol. You won't have any weird side effect, but it just won't work. Which means that you have to start taking the complete course of antibiotics again. But before you take them again you have to wait a while....so it will be like a month or so before you get any better.
And once you start taking them, you must take them at regular intervals, and ALL of them. Even if you feel better half way through, you should ALWAYS finish the course or it will come back.

Why not just don't drink while your sick, any you'll get better faster, well actualy you'd get better at the normal rate the alcohol can slow recovery times and yes it can make antibiotics ineffective, Do ya gotta have it that bad, hell I been there myself, just a beer or glass of wine or two in a day should not harm ya that bad, but obviously dude, come on, if you get **** faced while your taking antibodies for a flu/virus etc.... what the hell do think will happen, come on ya'll, ya killing me....lol
