It doesn't matter if it's his first time being bred or not, nor does it matter in humans, or anything else for that matter. If you keep them together for at least two of her heat cycles then she'll most likely be pregnant. She could become pregnant their first night together during her heat cycle. You need to take the male out though once she does become pregnant.
Also, do you have the proper knowledge to breed? Do you have homes lined up for the babies or are you planning on keeping them yourself? Make sure you're breeding responsibly.

WHY ARE YOU BREEDING THEM AT ALL?

Look on www.petfinder.com. Do a search for guinea pigs and see how many are homeless. Do you think this is a money maker or something? I currently have 30 guinea pigs in my rescue and I turn people away all the time because I am full! I am one rescue. It is because of people like you who randomly breed their guinea pigs for fun or because they will make money and then cannot find anyone to take them.

Then they put a sign up and say free guinea pigs to good home which get snatched up by people with snakes or monitor lizards. Or, better yet, fighting dogs!

I am sure you don't even know when the babies are sexually mature so then the mama will get knocked up by the babies OR the babies will knock each other up which can cause a whole host of other health issues.

It is people like you who make me sick and for why there are rescues like mine that have to exist; that have to clean up after you screw up or get tired of your little venture and decided you don't want to do it anymore.

There is a special place in the underworld for people like you...

Probably...but let them go at it a few times...never hurts to be sure....and I'm sure he wouldn't mind doing it.
