I know that yellow cake mix includes yellow food coloring. If I made a yellow cake from scratch, what would make it yellow? Is there a significance to the yellow-ness, or did someone just say "I think yellow cake would look prettier than white cake"?

The egg yoke makes is yellow. White cake usually has less or no egg yoke. I've always thought white cakes were fluffier, but it all depends on the recipe and perception. I know nothing about the science, just the taste.

Whole eggs make it yellow, white cake only uses egg whites.

As JayJay said, It is the egg yolks. White cakes usually only have egg whites in them.

Possibly the yellow color could come from butter and/or lemon ingredients.
