Yes you can. 65 is the international direct dialing (IDD) code for Singapore. If you're calling from a mobile, add + before the 65

65 is Singapore's postal code. I assume 68545946 is the high com's telephone number. You don't need to dial an area code because Singapore is a small country, we don't use area code here.
