A certain amount of energy is lost to the auxilliaries such as the water pump, fuel pump, oil pump and fan. In general for a two cylinder engine, each rotating part (cam, crankshaft, ect.) there are three bearings for part.
With four cylinders, you add little if any energy loss because of the auxilliaries and you will probably only add one additional bearing for cylinder.
The result should be a very very slight increase in efficiency.

as u keep on increasing the cyclinder, the Power of the Vehicle increases but it consumes more fuel..
thats why a motorbike is more Efficient than a Sedan
but Fuel efficiency can also be defined as Total power developed per liter of Fuel , then
increasing the cyclinder help in better torque management (there is lot fluctuation of torque in a single cyclinder , )hence fuel effiency increases if u increase the cyclinder...

it depends on the efficiency of design of the whole motor... not the number of cylinders ...so, the fuel management system, the cam lift and duration, the shape and efficiency of the combustion chamber, size and stroke of the pistons...
