I think this answer depends on what type of trip you are taking.

For quick last minute deals I use priceline, mostly for hotels. You can get really really cheap 4 star hotels on there.

I recently went on a cross country road trip and AAA was the best place to go. Definately sign up if you're not a member. They can provide more detailed road directions than mapquest - using the Triptik service. It even tells you where road construction/traffic is. They have books on camping, resorts like Disney, and they have really friendly and helpful staff no matter where you go across the country. All the materials are free if you're a member. I would recommend them for domestic/caribbean travel help, if you'd rather work with someone one-on-one rather than online.

For overseas travel I would highly suggest that you use a travel agent, unless you are visiting relatives/friends who can assist you. There are often special requirements, departure fees, taxes, and other miscellaneous fees to visit various countries. A travel agent would have the latest information on all these things. They can also assist with travel insurance. If you're shelling out a lot, it is best to use a professional. Agents are also good if you have no idea where you want to go, or are looking for a deal overseas. Make sure to choose an agent that has a good reputation in the area.

For an interesting tour experience try local community groups that sponsor trips. These are usually well planned and often include meals. Tour directors plan these trips to see the most sights and for the best deal. These tours are usually for adults or seniors. Check your local newspaper's travel section.

Finally if you know exactly where you want to go and feel comfortable organizing your trip then the internet is the way to go. I've heard good things about kayak.com which searches all the discount sites. Do your research though. There are differences in prices on the discount sites. Also watch the trip cancellation policies on the sites as well. Priceline and hotwire allow you to name your own price if airplane arrival and depature dates are flexible. I recommend bidding at least 50% less than what the other sites are showing you. Don't book airline tickets off airline sites. They are wayyyy more expensive.
