I think it is a sign that a person doesn't know who they are in God. I struggled with it for years. It wasn't until I realized who I am in God and that I have a purpose that my low self esteem was broken. I am somebody in Christ.

No. Having low self-esteem is not a mortal sin. Are you Catholic? Just wondering since Catholicism is the only mainstream religion I can think of that actually differentiates types of sin using particular labels (i.e. mortal sin vs. venial sin).
If you Catholic (and even if you're not) remember that mortal sin is a sin that 1) is utterly terrible; 2) you commit with full knowledge; and 3) you commit deliberately (knowing it's wrong). Having a low self-esteem doesn't meet this definition.
You should talk to someone about why you feel this way. Did someone tell you that having a low self-esteem is a mortal sin? If so, they're wrong. God loves you. You should figure out how to allow you to love yourself. Good luck!

Why did you use the term "mortal sin"? According to the Bible, all sin is deserving of death.
-It says in Romans 6:23 "For the wages of sin is death; but the gift of God is eternal life through Jesus Christ our Lord."

Not generally, but if taken to far I think it would be. Having doubts in yourself is not sinful. Having doubts that God can do the things through you is. Anything is possible with God.
