The second time that God told Moses to bring water out of a rock, he told Moses to speak to the rock. Moses instead hit it with his staff.
That may not sound like such a big deal, but let me break it down. Moses disobeyed a DIRECT order from God, and he also put more faith into the staff than he did in God's authority. (Remember, Moses' staff had turned into a serpent and back, and it had also brought water out of a rock, so it kinda became like a "magic staff" for Moses, and he forgot that it was God who made the staff do those things.)

Because at the second rock he was only to speak to it, and instead he was mad at the people for blaming him for their lack of water, and he struck the rock instead. His anger at the people was a selfish anger, as opposed to the anger he felt when he came down off the mountain and they were worshiping the calf. His anger at that time was against the people for God, instead of against the people for himself.
