Tell me something about yourself is an open ended question to get the interviewee ( you) talking. This would usually form the basis of verifying
a) Soundness of your thought process ( not thoughts)
b) Integrity
c) How you evaluate yourself
d) Your perspective of the things around you.
Based on the situation whether its a interview for a job or a tv show, highlight the relevant areas first ( preferably chronologically - data wise).
Highlight your achievements in these and learnings from your failures ( remember.. the person who learns from failure , will not fail the next time so is an asset!)
Also its advisable to divide the answer into sections for eg.. on the Professional front < answer xyz > , on my educational front < answer abc> , on my family / personal front < blah blah>.
Try to be precise ( not verbose). Prepare some smart statements to give a higher recall of your interview. ( for eg, for a tech support job.. I take calls / solve tickets is strict NO. " I solve problems " is more positive thing).
If you can, this is an ideal point to lead an interviewer to a territory familiar to yourself. Use it!

What the interviewer is looking for is information on how you can best support their business. It is always a good idea to research the business that wants to interview you. Tell them whatever is true about yourself that fits with their mission statement, if they have one, or the job description. Tell them how wonderful you are; sounds silly but you must sell yourself proudly. Here is a Web site with sample Q&A's.

Be concise. Be short for 1 minute (atmost). If you are fresher, start with schooling, any acheivements on the way (academic wise if u attending for white collar job, or extra-curricular, if it suits your target job). Then college, year of passing, again acheivements (if any). Any special skill that will bring benefit to the job (any foreign language proficiency or co-ordination activities). Then few really TRUE positive attitutes of yours. And close it with why you want to be in that field and why you have applied for that company (Do a homework, know abt the company before you go there). Any part-time job experiences may help. Basically, they dont wanna hear the "facts" about you, they are looking for someone who suits them. So cleverly put thru few acheivements or your special skills ..and what benefits you can bring in (apart from technical skills). It is this one minute you have got to stand out from crowd.
For experienced ppl. they can start with college degrees, any certifications, previous companies, promotions, acheivements, anything they did that brought something good for thier companies (be it business or customer relations etc). Targets set/acheived ..types of projects worked on etc.. it varies based on type of job but the idea is same. Project yourself that you are worthy enough to be hired. Put ur positive points forward and make them start thinking "hey if i recruit this guy, he will bring something extra to his skillset". Surely interviewer will take it from there and will start asking you specific questions so be careful about the words you put in your first minute and have all possible answers (and expected questions) ready.

First introduce yourself.Your education,experience and skills.Prepare yourselves in such a way to answer some frequently asked questions by consulting some persons who have attended interviews recently.
Briefly explain your background.
Go to a library and search books on "Interviews" for more informations.

I usually give a very brief background of what kind of work I've been doing together with my educational background.
I rarely tell them anything about my personal life unless they ask. That being said, there is nothing wrong with inserting something like "I play in a volleyball league and like to play squash."

the answer should give a fair idea to the interviewer about you.
Give a brief background of your family. Your academic history (where you studied what). Your work experience. Your interests and hobbies etc,.
Keep it short and sweet, dont get into too many details...
