I was working on an old house the other day, that no one is living in and we are trying to fix up to sell, I had jeans on. I'm 25 and female, not that this matters. I was sitting cross legged off and on changing doorknobs on the house, and i told my husband that my leg hurt. (in general).... thought my knee and behind my knee was hurting because I was getting up and down and cross legged. Anyway, I ran my hand up and down my thigh and calf and felt a gumball sized ball behind my knee. I dropped my pants, and saw what looked like a single puncture wound, small,a nd the area around it was red and inflammed. It does not/didn't itch at all. Just hurts. It's right in the crease of my leg. Now, 36 hours later, it still hurts like hell, is red, and inflammed. the puncture is darker. and i'm limping, mainly because the pain is in the crease and it presses when i walk. i didn't see what bit/stung me, but it did not hurt when it happened. it hurt after. what do you think? i'm in pain!

Actually it doesn't sound like a bite to me. It seems more likely that something punctured your skin either through your jeans or it was already inside your jeans when you put them on and then when your knee folded, the object was forced into you.
You need to clean the area; soak a cloth in perooxide and let it sit on the wound. After about five minutes, remove the cloth and have your husband look to see if he can see any foreign object in the wound. If he cannot, or cannot remove an object he does see, you should probably consult a doctor. If you don't know how to maintain a sterile field, you should probably see a doctor rather than having your husband tend to you.

It could be almost anything, wood, glass, plastic, metal. Since it is in an area that requires constant contact, it should be removed.

Go to the ER. Don't neglect a bite from an unknown creature. It took 6 months for some kind of bite on my foot from a tropical insect to heal and several trips to a dermatologist.

I highly recommend Wellman's Peroxide Ointment.
It heals most types of poisonous stings in a jiffy.
For stopping of bleeding instantly, I recommend Hydrogen Peroxide. These are trully effective and well tested by my own experience to be brief.

could be a spider bite do you live in the area with the brown recluse?if it keeps growing in size and hurts call the doctor!
