Is this sentence gramatically incorrect?

"How many brothers have you got?"

If it's grammatically incorrect, are there some other ways to say "How many brothers do you have?"

Both are correct, but teachers prefer to hear "How many brothers do you have?"

Some people seem to forget that nowadays English is spoken all over the world, in lots of different countries and by millions of people.
And NOT always the same rules apply, and different vocabulary or structures are used, depending where you are.
Both your sentences are correct, but I would say that, in Europe at least, the question "how many brothers (and sisters) have you got" is more common or more frequently used.

You should not end a sentence with "got" or "have." Perhaps use family or something to that effect. Maybe ask, How many brothers/male siblings do yu have in your family?
I think the other way sounds as if the sentence is incomplete.

I guess the easiest way to start is to say that "got" is the past tense of "get" (to fetch, to obtain, to receive, seize, propagate, understand; it is the act of coming in the state of having, becoming the owner or receiving, obtain, acquiring, etc.)

The difference with "have" is very subtle. Have is to hold in the hand or control or own, possess as part, charcterististic or attribute (I have black eyes is not the same as "I got black eyes," which indicates that you got punched or hurt, and not the same as when the color of your eyes are black, and not blue or hazel).

"I got an apple," means that I went and fetched an apple, and "I have an apple" means that it is in my possession.
