It's usually more of a concern for a muffin to be stale rather than to be mouldy. Fresh muffins keep well for about five days (longer if you freeze them).

You can smell mold. Mold grows in warm, moist environments and can appear white, black, grey, or green in color (sometimes it looks fuzzy). I doubt a freshly made muffin would have mold on it.

when it starts to mold..youll be able to tell..trust me..the smell will clue you in too..that and it will be allot more moist when touch it.
