You can lock it w/ a special TSA lock that you can get at most travel stores and luggage section in department stores. These locks are then only openable by you and the TSA inspection folks who have a skeleton key for these locks. They sometimes open it and look through it, so they need to be able to open it.

I'm pretty sure they just throw it through the big scanning device. It should be safe to lock it. They should be able to get what they need just by the screening.

We were told not to put locks on and had sheets in 2 of our suitcases that they had been opened.

You can only lock your checked luggage if you use a TSA approved lock which they can open.
